Genetic characterization of Thrace honey bee populations of Turkey: restriction and sequencing of inter cytochrome C oxidase I-II (CoxI-CoxII) genes

In this study, the genetic characterization of Thrace honey bee populations of Turkey were identified using the mitochondrial gene region, 3 end of cytochrome C oxidase I gene to 5 end of cytochrome C oxidase II gene (inter CoxI-CoxII genes) by using XbaI PCR-RFLP and DNA sequencing. A total of 322 worker honey bees were collected from five different localities; 4 provinces (Tekirda, Krklareli, Edirne and Canakkale) and Gokceada Island near Thrace. PCR RFLP and sequence data combined revealed three haplotypes. The most frequently observed haplotype (Type 1), revealed only one XbaI restriction site in inter CoxI-CoxII genes. Type 2 haplotype revealed additional XbaI restriction site and this digestion revealed three bands. The rarely seen third haplotype, which is first reported in this study, was only found in two of the samples that were collected from Gokceada Island. According to Maximum Likelihood phylogenetic analysis which was constructed on the basis of the distance matrix, Type 1 haplotype was clustered in the same clade with A. m. caucasica and A. m. anatoliaca whereas Type 2 was clustered in the same clade with A. m. macedonica and A. m. carnica. These results may indicate that Carniolan or Macedonian honey bee subspecies or their ecotypes can be found in the Thrace region of Turkey, mainly in samples that were collected from Krklareli and Edirne provinces near the borders with Greece and Bulgaria. En este estudio, la caracterizacion genetica de las poblaciones de abejas meliferas de Tracia en Turquia se realizo utilizando la region del gen mitocondrial comprendida entre los genes CoxI y CoxII, mediante el uso de PCR-RFLP con la enzima XbaI y la secuenciacion de ADN. Se recogieron un total de 322 abejas obreras de 5 localidades diferentes: 4 provincias (Tekirda, Krklareli, Edirne y Canakkale) y la isla de Gokceada, cerca de Tracia. La combinacion de PCR-RFLP y la secuenciacion revelaron tres haplotipos. El haplotipo mas frecuentemente observado (Tipo 1), mostro solo un sitio de restriccion con XbaI entre los genes CoxI-CoxII. El haplotipo tipo 2 mostro un sitio adicional de restriccion con XbaI y esta digestion manifesto tres fragmentos. El tercer haplotipo poco frecuente, que se describe por primera vez en este estudio, solo se encontro en dos de las muestras recogidas en la isla de Gokceada. De acuerdo con el analisis filogenetico del UPGMA, que se construyo sobre la base de la matriz de distancia, el haplotipo tipo 1 se agrupo en el mismo grupo con A. m. caucasica y A. m. anatoliaca, mientras que el tipo 2 se agrupo en el mismo clado que A. m. macedonica y A. m. carnica. Estos resultados pueden indicar que las subespecies de abejas meliferas carniolas o macedonicas o sus ecotipos se encuentran en la region de Tracia (Turquia), principalmente en las muestras recogidas en las provincias de Krklareli y Edirne, cerca de las fronteras con Grecia y Bulgaria.
