The EtOAc extract ofC. italicumwas purified by different chromatographic methods. Compound1was isolated from the whole plant ofC. italicum. Its structure was established on the basis of UV, IR, 1 D, 2 D NMR, and HRESI-MS methods and antioxidant activities of this compound were investigated using TEAC, FRAP, and CUPRAC assays. According to the ABTS assays, the TEAC capacity of compound 1 was determined as 0.186 mmol TE/g. [GRAPHICS] .Öğe Accumulation of essential and toxic metals in sediment from the marmara sea along tekirdağ coast: Risk assessment for ecological health(Desalination Publications, 2019) Dökmeci, Ayşe Handan; Şabudak, Temine; Dalmış, V.In our study, sediment samples were collected from four different stations in the spring of 2018 to reveal the presence of essential and toxic metals in the Marmara Sea along the coast of Tekirdağ. Concentrations of essential and toxic metals in sediments of Cd, Ni, Cu, Pb, Cr, Mn, Hg, Fe, As and Zn were measured with an inductively coupled plasma optical emission spectroscope device. The mean essential and toxic metal contents of sediment samples were determined as Fe (10,646.65 mg/kg) > Mn (265.03 mg/kg) > Ni 20.46 mg/kg) > Zn (18.63 mg/kg) > Cr (16.34 mg/kg) > As (11.94 mg/kg) > Pb (5.22 mg/kg) > Cu (3.8 mg/kg) > Cd (2.3 mg/kg). Hg could not be determined because it is in the below detection limits. The potential ecological risk index was calculated for all the metals, except for Hg, in four locations. In the sample SDM1 considerable risk for Cd was detected, moderate risk was found in SDM2 sample and low risk was detected in SDM3 and SDM4 samples. No other ecological risk was found for other metals. The reason for the very high ecological risk was calculated for cadmium might be that cadmium has high toxicity coefficient compared to other metals and due to the use of phosphorous fertilizer containing cadmium in agricultural activities and activities in the field of industry affecting natural water flows, erosion or discharges passing through the Marmara Sea along the coast of Tekirdag province. © 2019 Desalination Publications. Bu amaçla; Alyssum sibiricum sırasıyla hekzan, kloroform, etil asetat ve metanol ile ekstrakte edilmiş ve bu ekstrelerde antioksidan aktivite; DPPH, ABTS, ?-Karoten-linoleik asit renk giderim aktivitesi ve CUPRAC yöntemleri kullanılarak araştırılmıştır. Antioksidan aktivite sonuçlarına göre; tüm yöntemlerde, en yüksek antioksidan aktiviteyi metanol ekstresi göstermiştir. A. sibiricum bitkisinin hekzan ekstresinin GC-MS sonuçlarına göre, 41 bileşik tespit edilmiştir. Bu bileşikler arasında; n-Dokosan (%23.24), palmitik asit (%15.02), laurik asit (%7.67) ve 1-hekzadekanol (%5.82), hekzan ekstresinin temel bileşiklerini oluşturmuştur.Öğe An estimation of the transfer of micronutrient (Cu, Mn, Zn) and toxic elements (Cd, Cr, Ni, Pb) From soil to rice (Oryza Sativa) in Evros-Ergene river basin(Parlar Scientific Publications (P S P), 2017) Dökmeci, Ayşe Handan; Şabudak, TemineIn this study, various concentrations of nonessential toxic heavy metals (Cd, Cr, Ni and Pb) and the micronutrients (Cu, Zn, Mn) in twenty brown rice (O.sativa) and relevant paddy soil samples were investigated. The soil and brown rice samples from Evros-Ergene river basin were extracted for their total heavy metals content by dry digestion method and then determined by using ICP OES. The results of this study reveal that the concentrations of Cd, Cr and Ni in the soil of the studied sites are above the maximum permissible levels. The average concentration of all metals in brown rice samples is below the maximum permissible levels of the WHO guideline. The ranking order of bioaccumulation factor (BAF) for heavy metals is Zn > Cu > Mn > Ni> Cr = Cd = Pb, which indicaties that the accumulation of micronutrients was more than that of nonessential toxic heavy metals. It can be concluded that these rice samples cannot be regarded as a complete source to determine all metals posing threat to human health.Öğe Antiinflammatory and antioxidant activities of Trifolium resupinatum var. microcephalum extracts(Asian Journal Of Chemistry, 2008) Şabudak, Temine; Dökmeci, D.; Özyiğit, Filiz; Işık, E.; Aydoğdu, NurettinThe antiinflammatory and antioxidant activities of the ethanol extract of Ttifolium resupinatum L. var. microcephalum (Leguminosae) have been assessed on arthritic rats. The extract (1.35 and 13.5 mg/kg, i.p.) significantly reduced the paw edema induced by the complete Freund's adjuvant. Furthermore, it also inhibited lipid peroxidation. These effects of T resupinatum extract may be directly linked to the content in triterpene saponins and flavonoid compounds and consequently to their free radical scavenging activities.Öğe Antioxidant Activity of Extracts of Soursop (Annona muricata L.) Leaves, Fruit Pulps, Peels, and Seeds(Inst Animal Reproduction & Food Research Polish Acad Sciences Olsztyn, 2019) Orak, Hakime Hülya; Bahrisefit, İlayda Şevik; Şabudak, TemineThe total phenolic content (TPC), total flavonoid content (TFC), and the antioxidant activity of soursop (Annona muricata L.) leaf, fruit pulp, seed, and peel extracts obtained using successive extraction with hexane (Hxn), dichloromethane (DCM), ethyl acetate (EtOAc), and methanol (MeOH) were determined. The Hxn soursop seed extract was analysed by GC-MS. The highest TPC was determined in MeOH extracts. MeOH and EtOAc extracts were rich sources of flavonoids. Generally, soursop leaf and fruit pulp extracts had the highest and the lowest both TPC and TFC, respectively. Fatty acids were dominant in the Hxn seed extract. Among antioxidants, terpenoids ((E)-nerolidolas as dominant) and phytosterols ((3-beta)-stigmast-5-en-3-ol with high content) were identified. The soursop seed, followed by leaf and peel extracts (MeOH and EtOAc) had the highest DPPH center dot scavenging activity, TEAC, FRAP, and CUPRAC. Antioxidant activity of peel extracts (MeOH and EtOAc) was particularly high in beta-carotene-linoleic acid emulsion system. Strong correlations were found between TPC, TFC, TEAC, FRAP, and results of DPPH assay. In conclusion, soursop leaves and fruit seeds and peels, which are cheap, waste plant material, could be considered as a source of phenolic antioxidants with a high antioxidant activity.Öğe Antioxidant activity of five new phenolic compounds from Cirsium creticum subsp. creticum(Elsevier Science Bv, 2019) Şabudak, Temine; Özer, Merve; Orak, Hakime Hülya; Çalışkan, HilmicanIn this study, five new alkyl catechol derivatives, 1, 2, 3, 4 and 5 were isolated for the first time from Cirsium creticum subsp. creticum L. and their structures were determined by using spectroscopic techniques, including mass spectrometry, 1D and 2D NMR. All of the isolated compounds were primarily evaluated for antioxidant activity against BHA and tocopherol. All compounds which were isolated from C. creticum subsp. creticum exhibited antioxidant activity, however it was noticed that the presence of a pentose sugar increased the antioxidant activity.Öğe Antioxidant and Tyrosinase Inhibitory Activities of Flavonoids from Trifolium nigrescens Subsp petrisavi(Amer Chemical Soc, 2013) Demirkiran, Özlem; Şabudak, Temine; Öztürk, Mehmet; Topçu, GülactiTrifolium nigrescens was researched for its chemical constituents for the first time. Bioassay-guided isolation of the EtOAc extract from the leaves of T. nigrescens resulted in the purification of a new biflavone, 4',5,5 '',7,7 ''-pentahydroxy-3',3'-dimethoxy-3-O-beta-D-glucosyl-3 '',4'-O-biflavone (1) along with eleven known compounds consisting of three phenolics (2-4), and eight flavonoid glycosides (5-12). Their structures were determined by extensive ID and 2D NMR, and MS data analyses. The isolated compounds were evaluated for their antioxidant activity and inhibitory activity on mushroom tyrosinase. Highly potent inhibitions Were found by compounds 7 (IC50 = 0.38 mM), 8 (IC50 = 0.19 mM), and 12 (IC50 = 0.26 mM) when compared with standard tyrosinase inhibitors kojic acid (IC50 = 0.67 mM) and L-mimosine (IC50 = 0.64 mM). The antioxidative effect of the isolated compounds and the extracts were determined by using, beta-carotene-linoleic acid, DPPH center dot scavenging, ABTS(+center dot) scavenging, superoxide scavenging, and CUPRAC assays. The experimental findings indicated that all the compounds demonstrated activity in all antioxidant activity tests employed except for the compounds 2-6. Compounds 2-6 showed moderate activity only in ABTS(+center dot) scavenging assay. The new compound 1 exhibited better activity than standard alpha-tocopherol in DPPH center dot scavenging, and ABTS(+center dot) scavenging assays. The results show that T. nigrescens can be regarded as a potential source of antioxidant compounds and tyrosinase inhibitors of significance in both the pharmaceutical and food industries.Öğe Assessment of heavy metal contamination in soil and wheat (Triticum Aestivum L.) plant around the çorlu-çerkezkoy highway in thrace region(Global Network Environmental Science & Technology, 2012) Ekmekyapar, Füsun; Şabudak, Temine; Seren, Gamze YıldızPollution caused by traffic activities is increasingly becoming a great threat to human health in the region of Thrace in north-west of Turkey. Thirty six soil and plant samples were collected from the vicinity of Corlu-Cerkezkoy Highway. The samples were taken at distances of, 1, 25, 50, 100, 250 and 500 m from the highway. The each plant sample was divided into two subsamples. One subsamples of each plant was washed with with distilled water, while other part was not washed and were analyzed without any washing. All samples were analyzed for their heavy metal concentrations (Pb, Zn, Cd, Cu, Ni, Mn, Cr and Fe). The results showed that the soils in the study area were polluted by Pb. The lead concentration in the soil showed values from 19 +/- 0.2 mg kg(-1) to 351 +/- 0.3 mg kg(-1) and found to be higher than the allowable maximum limits in soils. The manganese levels of soils were slightly high and varied from 182 +/- 0.1 mg kg(-1) to 806 +/- 0.1 mg kg(-1). In this study, metal concentrations of unwashed plant samples were higher than those of the washed ones. This study indicated that the heavy metal contents decreased with increasing distances from the highway in unwashed plants.Öğe Bioaccumulation of essential and toxic metals in four different species of bottom fish in the marmara sea, tekirdag, Turkey: Risk assessment to human health(Desalination Publications, 2019) Dökmeci, Ayşe Handan; Şabudak, Temine; Dalmış, V.Due to the intensive industrial and agricultural activities in the Thrace region and the maritime transport that directly and/or indirectly affect Tekirdağ coastal areas, the pollution starts from the Tuna River and extends to the Marmara Sea through the Black Sea and the Bosphorus. Heavy metals are not only a threat to marine life, but also to the health of people who consume seafood. In order to investigate the presence of essential and toxic metals in Marmara Sea fish species, samples of 4 different bottom fish (sole, whiting, striped red mullet and angler fish) were collected from 4 different stations in the coastal line of Tekirdağ province in the spring of 2018. Bioaccumulation of essential and toxic metals (Cd, Ni, Cu, Pb, Cr, Mn, Hg, Fe, As, Zn) in the muscles of fish samples was measured by ICP-OES instrument. The accumulation pattern of the analyzed metals in all bottom fish muscles was as follows: Fe (58.8) > Zn (23.9) > Mn (3.3) > As (1.44) > Cu (0.8) > Cr (0.6) > Ni (0.3) > Cd (0.08) mg/kg dry weight. ANOVA analyses indicated significant variations (p < 0.05) of Fe, Zn, Mn, As, Cu, Cr, Ni, and Cd in the muscles of different fish species. Significant variations were observed in different stations (p < 0.05), and As, Pb and Hg were below the detection limit of ICP-OES. Except Cu, the levels of all essential and toxic metals in fish samples were found to be above the national and international legal limits recommended for human consumption. Since the THQ values of inorganic As were higher than 1 mg/kg d·w, it is likely to pose health risk for the consumers of the these fish species. © 2019 Desalination Publications.Öğe BIOACTIVE COMPOUNDS, ANTIBACTERIAL AND ANTIFUNGAL ACTIVITIES OF TWO Cirsium SPECIES(Wydawnictwo Akad Rolniczej W Lublinie, 2019) Gülen, Dumrul; Şabudak, Temine; Orak, Hakime Hülya; Çalışkan, Hilmican; Özer, MerveIn the GC-MS analysis, 81 bioactive phytochemical compounds were identified in hexane extracts of Cirsium creticum and Cirsium italicum. Terpenoids constituted the main fractions of C. italicum (70.95%), while hydrocarbons were most abundant compounds of C. creticum (41.11%). The antibacterial activity and antifungal activity of extracts from two Cirsium species was tested using microdilution methods. According to the results of antibacterial activity, the highest inhibition effect of C. creticum was found on B. subtilis, E. coli and P. aeruginosa. The highest inhibition effect of C. italicum was found on B. subtilis. In the results of antifungal activity, the highest inhibition effect of C. creticum was found on P. chrysogenum and the highest inhibition effect of C. italicum was found on C. krusei. The present work is the first report on hexane extracts compounds of two Cirsium species as well as antibacterial and antifungal activities.Öğe Biodiesel production from waste frying oils and its quality control(Pergamon-Elsevier Science Ltd, 2010) Şabudak, Temine; Yıldız, MelahatThe use of biodiesel as fuel from alternative sources has increased considerably over recent years, affording numerous environmental benefits. Biodiesel an alternative fuel for diesel engines is produced from renewable sources such as vegetable oils or animal fats. However, the high costs implicated in marketing biodiesel constitute a major obstacle. To this regard therefore, the use of waste frying oils (WFO) should produce a marked reduction in the cost of biodiesel due to the ready availability of WFO at a relatively low price. In the present study waste frying oils collected from several McDonald's restaurants in Istanbul, were used to produce biodiesel. Biodiesel from WFO was prepared by means of three different transesterification processes: a one-step base-catalyzed, a two-step base-catalyzed and a two-step acid-catalyzed transesterification followed by base transesterification. No detailed previous studies providing information for a two-step acid-catalyzed transesterification followed by a base (CH3ONa) transesterification are present in literature. Each reaction was allowed to take place with and without tetrahydrofuran added as a co-solvent. Following production, three different procedures; washing with distilled water, dry wash with magnesol and using ion-exchange resin were applied to purify biodiesel and the best outcome determined. The biodiesel obtained to verify compliance with the European Standard 14214 (EN 14214), which also corresponds to Turkish Biodiesel Standards. (C) 2010 Elsevier Ltd. Two new biflavonoids, 5,5'',7''-trihydroxy-3,7-dimethoxy-4'-4'''-O-biflavone (3), (8,3',4',5'-tetrahydroxy-7-O-alpha-glucosyl isoflavone)-6-8''-(4'''-hydroxy-7''-O-alpha-glucosyl isoflavone) biflavone (7), a new isoflavone, 6-2'-dihydroxy-5-methoxy isoflavonone-4'-O-alpha-D-glucopyranoside (5) and a new phenolic compound, ethyl-(2,3-dihydroxy-5,6-dimethoxy-4-acetyl) benzoate (4), together with three known compounds (1), (2) and (6) were isolated from Cirsium italicum. Their structures were elucidated by extensive 1D and 2D NMR, and MS data analyses. All compounds isolated from C. italicum exhibited antioxidant activity. However, the highest activity of the EtOAc extract is considered to be responsible for compounds (1), (2) and (3) being the main compounds. There are several different ways that heavy metal pollution ends up in our oceans. lakes and bays. The four main ways are: burning heavy metal, heavy metal runoff, dumping or heavy metals, and tributary inflow. Roadways and automobiles are also considered to be one of the largest sources of heavy metals. Zinc, copper, and lead are three of the most common heavy metals released from road travel, accounting for at least 90 of the total metals in road runoff. Meanwhile. they are natural components of the Earth's crust. They can not be degraded or destroyed. To a small extent they enter Our bodies via food, drinking water and air. Heavy metals become dangerous when they tend to bioaccumulate. Since study area has been Under a heavy industrialisation period, alteration of natural water resources has been reported in the area. In order to evaluate the potential of this alteration heavy metal monitoring program was run in the area. In the region, drinking water is supplied by groundwater abstraction. Surface water has limited usage because of its low quality. Surface water has both organic and inorganic based Pollution related to uncontrolled agricultural and industrial activities. Both groundwater and Surface water samples were collected from over 30 sampling points. Cadmium and copper analyses were carried out via atomic absorption spectroscopy (AAS). Great variations in the results among the samples were recognised during analysis. Maximum concentrations for both cadmium and copper were determined in surface water samples as 202 ppb and 19 ppb, respectively. In the study area industrialisation has been growing rapidly and uncontrolled since 1970's. Corlu has a great importance regarding its industrial improvement both for Thrace region and Turkey. This consequence has not yet been evaluated sufficiently in the view of environmental concerns. Due to environmental cycle, trace elements, emanating from atmospheric and industrial Pollution accumulate, in soil and affect the ecosystem nearby. These elements may accumulate in soil, collected in plant or infiltrated 10 ground water. If they accumulate in a plant the), may reach to food chain and become harmful for human life. Hence, the investigation of trace elements in soil and plant samples is very important in the point of environmental Pollution, especially for plants. In the present study, levels of Pb and Ni in soil and plant samples collected from Corlu, Turkey, were determined by flame atomic absorption spectrometry (FAAS). The maximum levels of trace elements were determined as 52.91 mg Pb kg(-1) and 40.68 mg Ni kg(-1) in soil and 80.90 mg Pb kg(-1) in plant parts. Phytotoxic Pb amount ill the some samples was observed. The nickel in plant samples was not in detectable limits for FAAS.Öğe Farklı Ayçiçeği Genotiplerinin Perikarp ve Tohumlarında Bulunan Fungal Etmenler ile Metabolitler Arasındaki İlişkiler Üzerinde Araştırmalar(2020) Özer, Nuray; Şabudak, TemineBu çalısmada, ayçiçeginde tohum kökenli fungal patojenlerle (Alternaria alternata, A. infectoria, Bipolarıs cynodontis, Cladosporium cladosporioides, Fusarium culmorum, F. oxysporum) farklı ayçiçegi genotiplerinin (mildiyö hastalıgına karsı tolerant; TTAE 1319, 13 TR009, TTAE-13-9, 11 TR015 CL, 13 TR 00 ve hassas; 2453 A, 9728 A, 9725 A, 2517 A, 9178 A) perikarp ve tohum kısımlarından üç farklı çözücü (hekzan, metanol, kloroform) ile elde edilen ekstraktlardaki bilesikler arasındaki iliskiler incelenmistir. Tüm genotiplerin tohumları perikarba göre daha yüksek miktarda metabolit içermis, kloroform en iyi çözücü olmustur. Tolerant genotiplerin tohumlarından kloroform ile elde edilen metabolik ekstraktlar tüm fungal etmenlere karsı en az bir kriter (koloni çapı, konidi çimlenmesi ve konidi çim tüpü uzunlugunda azalma, inhibisyon zonunda artıs) yönünden yüksek antifungal etki göstermistir. Etmenlerin genotip kısımlarında bulunma oranı ile en yüksek düzeyde pozitif iliski 3?- bromocholest-5-ene (steroid) ile B. cynodontis arasında gözlenmistir. 2-Dodecyl-dodecen (alken)?in miktarının artması ile A. alternata?nın çim tüpü uzunlugunda yüksek oranda azalma oldugu, octadec-9-enoic acid (yag asidi), bombykol (alkol), benzedrex (amin), hexadecanamide (amid), dodecyl cyclohexanecarboxylate (ester)in miktarının artısı ile sırasıyla A. infectoria, B. cynodontis, C. cladosporioides, F. culmorum, F. oxysporum?un konidi çimlenmesinde yüksek oranda azalma oldugu tespit edilmistir. Tüm bu bilgiler ayçiçeginde tohum kökenli fungal etmenlerin kontrolüne ve ıslah çalısmalarına katkıda bulunacaktır.Öğe Fatty acids and other lipid composition of five Trifolium species with antioxidant activity(Taylor & Francis Ltd, 2009) Şabudak, Temine; Öztürk, Mehmet; Gören, Ahmet C.; Kolak, Ufuk; Topçu, GülaçtıThe contents of fatty acids and other lipids of Trifolium balansae Boiss, Trifolium stellatum Lin., Trifolium nigrescens Viv. subsp. petrisavi (Clem) Holmboe, Trifolium constantinopolitanum Ser., and Trifolium resupinatum L. var. resupinatum L. (Leguminosae) were determined by gas chromatography-mass spectrometry (GC-MS). The whole plant hexane extracts of five Trifolium species contained eight fatty acids consisting of linolenic (31.1%) and palmitic (18.9%) acids as the most abundant unsaturated and saturated fatty acids, respectively. The total unsaturation for the oils of five Trifolium species was 30.6-42.2%. In addition to fatty acids, some alkanes and phytol were also obtained. Objective: We aimed the determination of the antioxidant and antibacterial activity of Cirsium creticum (Lam) d'Urv. subsp. creticum included Asteraceae family which is wild plant species in Trakya region. Methods: Crude n-hexane, diethyl ether, ethylacetate and methanol extracts from whole plants was used the determination of antiradical and antibacterial activity in C. creticum and compared in terms of extracts efficiency. Results: EC50 values of DPPH activity in different solvents revealed significant differences similar to TPC content. The lowest inhibitory effect was found to be Gram negative for E. coli, despite the highest inhibitory effect against Gram positive S. aureus. Conclusion: Antiradical and antimicrobial activity results presented that C. creticum, could be evaluate in food supplements and pharmaceutical industry as natural compound, and future investigations will be aimed the isolations of biologically active compounds to define individual active components.Öğe Investigation of Antioxidant Activity and Fatty Acid Contents of Solanum dulcamara(Springer, 2017) Makasçı Afacan, Ayşe; Şabudak, Temine; Ortaköy, Tarık; Kıyak, GizemThe fatty acid compositions of the leaves and fruit of Solanum dulcamara were determined by gas chromatography. The leaf oil of S. dulcamara is remarkably rich in linolenic acid. Among the S. dulcamara extracts, according to the 2,2-diphenyl-2-picrylhydrazyl method, hexane and methanol show the highest activity levels in fruit and leaves, respectively. According to the beta-carotene-linoleic acid method, dichloromethane has the highest activity level in both fruit and leaf. However, according to the modified cupric-reducing antioxidant capacity method, ethyl acetate and dichloromethane have the highest activity levels in fruit and leaves, respectively. The antioxidant activity levels and fatty acid contents of S. dulcamara are presented for the first time in this study.
