Korunga yem bitkisinde tohum kökenli Alternaria alternata'nın uçucu yağlar ile mücadele olanaklarının araştırılması

Özet
Bu çalışmada, korunga yem bitkisinde tohum çürüklüğü hastalığına neden olan Alternaria alternata (Fr.) Keissl.'ya karşı, kekik (Thymus vulgaris), okaliptus ( Eucaliptus globulus), defne (Laurus nobilis), mersin (Myrtus communis) ve adaçayı (Salvia officinalis) uçucu yağlarının in vitro koşullarda antifungal aktiviteleri araştırılmıştır. Bu amaçla in vitro koşullarda 1 µl/petri, 10 µl/petri, 50 µl/petri dozları uygulamaya alınmıştır. Uçucu yağlar otomatik mikro pipetlerle steril kültür antibiyogram disk kâğıtlarına damlatılmıştır. Parafilm ile kapatılan petri kapları 22±2 °C' de 8 gün boyunca inkübe edilmiştir İnkübasyon süresi tamamlandıktan sonra ölçümler yapılmış ve misel gelişiminin engellenmesi yüzde olarak belirlenmiştir. Bu araştırma sonunda, uçucu yağlardan kekik, okaliptus, defne mersin ve adaçayı 50 µl/petri dozlarında A. alternata 'ya karşı sırasıyla %100, %94,21, %84,92 %80,64 ve %43,46 oranında antifungal etki göstermişlerdir. Denemeye alınan tüm uçucu yağlarda 1 µl/petri, 10 µl/petri dozları arasında fungal patojenin baskı altına alınması yönünden herhangi bir fark olmadığı gözlemlenmiştir. Uçucu yağlardan kekik, okaliptus, mersin, defne ve adaçayı 1 µl/petri, 10 µl/petri dozlarında A. alternata 'ya karşı sırasıyla %99.80, %63.57, %58.78, %56.78 ve %37.68 oranlarında antifungal etki göstermişlerdir. Bu araştırmada doğaya dost uçucu yağlar ile in vivo çalışmalara ışık tutabilecek veriler elde edilmiştir.
In this study, antifungal activities of essential oils of thyme (Thymus vulgaris), eucalyptus (Eucaliptus globulus), laurel (Laurus nobilis), myrtle (Myrtus communis) and sage (Salvia officinalis) plants were investigated in vitro conditions against Alternaria alternata (Fr.) Keissl. For this purpose, doses of 1 µl/petri, 10 µl/petri, and 50 µl/petri were used in vitro conditions. Essential oils were dropped on sterile culture antibiogram disc papers with automatic micropipettes. Petri dishes sealed with parafilm were incubated for 8 days at 22±2 °C. Measurements were made after completion of the incubation period and percent inhibition of mycelial growth was determined. At the end of this study, essential oils of thyme, eucalyptus, laurel myrtle and sage showed 100%, 94.21%, 84.92% 80.64% and 43.46% antifungal effects against A. alternata at 50 µl/petri doses, respectively. It was observed that there was no difference in suppression of fungal the pathogen between 1 µl/petri and 10 µl/petri doses of all essential oils included in the experiment. The essential oils of thyme, eucalyptus, myrtle, laurel and sage showed antifungal effects at 1 µl/petri and 10 µl/petri doses against A. alternata at 99.80%, 63.57%, 58.78%, 56.78%, and 37.68% , respectively. In this research, data that can shed light on in vivo studies with nature-friendly essential oils were obtained.
