Tekirdağ ilindeki bağ üretim alanlarında Grapevine fanleaf virus (GFLV) ve Grapevine leaf roll associated-viruses(GLRV3-6)'lerinin saptanması üzerine araştırmalar

Özet
Tekirdağ ilindeki bağ üretim alanlarında verim ve kaliteyi olumsuz yönde etkileyen Grapevine fanleaf virus (GFLV) ve Grapevine leaf roll associated-viruses (GLRV3-6)'lerin saptanması amacıyla 2018 yılı Temmuz ve Ağustos aylarında sürvey çalışmaları gerçekleştirilmiştir. Sürvey çalışmalarında Tekirdağ ili merkez ilçe Süleymanpaşa'dan 105 yaprak örneği, Şarköy ilçesinden 146, Malkara ilçesinden ise 5 yaprak örneği olmak üzere toplam 256 adet simptom gösteren yaprak örnekleri toplanmıştır. Böylece Sürveyler esnasında yapraklarda kızarma, sarılık, mozaik, nekroz, yelpaze yaprak oluşumu, geriye doğru kıvrılma, şekil bozukluğu ve bodurluk simptomları gibi tipik virüs hastalıkları belirtileri gösteren yaprak örnekleri toplanmış ve bu örneklerde Grapevine fanleaf virus (GFLV), Grapevine leaf roll associated-virus 3 (GLRV-3) ve Grapevine leaf roll associated-virus 6 (GLRV-6) enfeksiyonları Double Antibody Sandwich Enzyme-Linked Immunosorbent Assay (DAS-ELISA) testi ile araştırılmıştır. Ayrıca GFLV'yi saptamak amacıyla Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) testi uygulanmıştır. DAS-ELISA test sonuçlarına göre 256 adet simptom gösteren yaprak örneğinden 60 adedinde GLRaV-3 (% 23.44), 7 adet örnekte ise GFLV saptanmıştır. 1 örnekte ise GLRaV-3 ve GFLV'nin karışık enfesiyonları belirlenmiştir. RT-PCR test sonuçlarına göre 256 örnekten 11 adedinin GFLV (% 4.3) ile enfekteli olduğu tespit edilmiştir. Şarköy ilçesindeki enfeksiyon oranı % 33.56 iken Süleymanpaşa ilçesindeki enfeksiyon oranı % 2.95 olarak belirlenmiştir. Malkara ilçesinden alınan 5 adet yaprak örneğinde GFLV, GLRaV-3 ve GLRaV-6 virüsleri saptanmamıştır. Toplam il düzeyindeki enfeksiyon oranı % 27.73 olarak tespit edilmiştir. Ancak GLRaV-6 virüsü testlenen örneklerin hiçbirinde saptanmamıştır.
The survey studies have been performed to identify Grapevine fanleaf virus (GFLV) and Grapevine ieaf roll associated-viruses (GLRV3-6), which have caused yield and quality losses in grapevine areas of Tekirdag province from July to August in 2018. During the survey studies, 256 symptomatic leaf samples, including 105 leaf samples in Süleymanpaşa district, 146 leaf samples in Şarköy district, and 5 leaf samples in Malkara district of Tekirdag were collected. Collected leaf samples exhibiting typical symptoms of virus diseases such as; reddish, yellowing, mosaic, necrose, fan leaf formation, rolling back, leaf deformation, and stunting symptoms were tested by Double Antibody Sandwich Enzyme-Linked Immunosorbent Assay (DAS-ELISA) method. Moreover, Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) method was applied to identify GFLV to samples. According to DAS-ELISA test, 60 leaf samples were found infected with GLRaV-3 (23.44%), and 7 samples had positive with GFLV from a total of 256 symptomatic leaf samples. However, one sample had mixed infection with GFLV and GLRaV-3. RT-PCR showed that the infection with GFLV of 11 leaf samples (% 4.3). While the infection rate in Şarköy district was 33.56%, the infection rate in Süleymanpaşa was determined as 20.95%. Five leaf samples collected from Malkara district were not found infected with GFLV, GLRaV-3 and GLRaV-6 viruses. The total infection rate in Tekirdag was 27.73%. However, none of the tested samples have been identified as GLRaV-6.
