Özet
Bu tez çalışmasında, Trakya bölgesinde yetişen Allium rumelicum, Jurinea kilaea, Peucedanum obtusifolium bitkilerinin toplam antioksidan aktivite ve kapasitelerinin, CUPRAC, ABTS/Persülfat ve DPPH yöntemleri ile analizlerinin yapılması planlanmaktadır. Ayrıca Allium rumelicum, Jurinea kilaea, Peucedanum obtusifolium bitkilerinin toplam fenolik ve flavonoid madde içeriklerinin ise Folin-Ciocalteu ve AlCl3/KAc yöntemleri kullanılarak belirlenmesi hedeflenmektedir. Bitkilerin antimikrobiyal özelliklerinin disk difüzyon yöntemi ile analizi sağlanacaktır. Bu çalışma sonucunda, CUPRAC ve ABTS/Persülfat yöntemleri ile analizlenen en yüksek toplam antioksidan kapasite/aktivite değerleri Jurinea kilaea bitkisinde gözlenmiştir. Aynı bitkinin DPPH yöntemiyle analiz edilen % inhibisyon değerinin en düşük olduğu tespit edilmiştir. Toplam fenolik madde içeriği tayininde ise Folin-Ciocalteu yöntemi kullanılmış ve burada da en yüksek gallik asit içeriğine sahip olan bitkinin Jurinea kilaea olduğu görülmüştür. AlCl3/KAc yöntemiyle toplam flavonoid içerik tayininde Peucedanum obtusifolium bitkisi en düşük kuarsetin içeriğine sahip iken, en yüksek kuarsetin içeriği Allium rumelicum bitkisinde saptanmıştır. Disk difüzyon yöntemi ile elde edilen antimikrobiyal aktivite analizi sonuçlarına göre; Peucedanum obtusifolium bitkisinin; üzerinde çalışılan 5 mikroorganizmadan 3'üne etkisinin olduğu belirlenmiştir. Jurinea kilaea bitkisi; üzerinde çalışılan mikroorganizmalardan sadece Salmonella enteritidis ATCC13076 üzerinde etkili olurken, Allium rumelicum bitkisinin ise üzerinde çalışılan mikroorganizmalardan sadece Staphylococcus aureus ATCC6538 üzerinde etkili olduğu tespit edilmiştir.
In this thesis, analysis of total antioxidant activities and capacities of Allium rumelicum, Jurinea kilaea, Peucedanum obtusifolium plants growing in Thrace region is planned with CUPRAC, ABTS/Persulfate and DPPH methods. In addition, it is aimed to determine the total phenolic and flavonoid substance contents of Allium rumelicum, Jurinea kilaea, Peucedanum obtusifolium plants using Folin-Ciocalteu and AlCl3/KAc methods. Analysis of the antimicrobial properties of plants will be provided by disc diffusion method. As a result of this study, the highest total antioxidant capacity/activity values analyzed by CUPRAC and ABTS/Persulfate methods were observed in the Jurinea kilaea plant. It was determined that the % inhibition value of the same plant analyzed by DPPH method was the lowest. In determining the total phenolic substance content, Folin-Ciocalteu method was used and it was found that the plant with the highest gallic acid content was Jurinea kilaea. In determining the total flavonoid content by AlCl3/KAc method, the Peucedanum obtusifolium plant had the lowest quarcetin content, while the highest quarcetin content was determined in the Allium rumelicum plant. According to the results of antimicrobial activity analysis obtained by disk diffusion method; Peucedanum obtusifolium plant; it has been determined that it has an effect on 3 of 5 microorganisms studied.
