Tekirdağ ilinde üretimi yapılan sert çekirdekli meyve ağaçlarında Plum pox virus (PPV) ve Prunus necrotic ringspot virus (PNRSV)'lerinin saptanması üzerine araştırmalar

Özet
Tekirdağ ili ve ilçelerindeki sert çekirdekli meyve üretimini olumsuz yönde etkileyen Plum pox virus (PPV) ve Prunus necrotic ringspot virus (PNRSV) hastalıklarını saptamak amacıyla 2018 yılı Haziran ve Temmuz aylarında sürvey çalışmaları gerçekleştirilmiştir. Sürvey çalışmalarında Tekirdağ ili merkez Süleymanpaşa ilçesinden 71 adet yaprak örneği, Şarköy ilçesinden 30, Muratlı ilçesinden 25, Çorlu ilçesinden 19 ve Malkara ilçesinden 13 adet yaprak örneği olmak üzere toplam 158 adet yaprak örneği toplanmıştır. Böylece mozaik, nekrotik ve klorotik lekeler, damar bantlaşması ve şekil bozuklukları sergileyen 76 adet kiraz, 35 adet erik, 28 adet şeftali, 19 adet kayısı yaprak örneklerinde PPV ve PNRSV'nin varlığı Double Antibody Sandwich Enzyme-Linked Immunosorbent Assay (DAS-ELISA) testi ile araştırılmıştır. Ayrıca halkalı leke ve damar bantlaşması simptomları sergileyen kiraz yaprak örneklerinde Plum pox virus (PPV), Reverse Transcriptase Polymerase Chain Reaction (RT-PCR) testi ile araştırılmıştır. DAS-ELISA testi sonuçlarına göre 16 adet örneğin PPV ile 4 adet örneğin ise PNRSV ile enfekteli olduğu tespit edilmiştir. RT-PCR test sonuçlarına göre kiraz yaprak örneklerinde PPV saptanmamıştır. Bu sonuçlar doğrultusunda %10.13 oranında PPV ile enfeksiyon oranı saptanırken, PNRSV ile enfeksiyon oranı %2.53 olarak tespit edilmiştir. Her iki virüsün neden olduğu ildeki toplam enfeksiyon oranı ise %12.66 olarak bulunmuştur.
The survey studies have been performed in June and July of 2018 to identify the Plum pox virus (PPV) and Prunus necrotic ringspot virus (PNRSV), negatively affecting stone fruit production in its districts and Tekirdag province. During the survey studies a total of 158 leaf samples, including 71 leaves from center district Süleymanpaşa, 25 samples from Muratlı district, 19 samples from Çorlu district, 13 samples from Malkara district were collected. Thus, symptomatic leaf samples exhibiting mosaic, necrotic and chlorotic local spots, vein banding, and leaf deformations symptoms of sweet cherry (76), plum (35), peach (28), apricot (19) were tested for the presence of PPV and PNRSV by Double Antibody Sandwich Enzyme-Linked Immunosorbent Assay (DAS-ELISA). Moreover, infected sweet cherry leaf samples showing ring spot and vein banding symptoms were tested by Reverse Transcriptase Polymerase Chain Reaction (RT-PCR). DAS-ELISA test results showed that the infections of PPV and PNRSV, on 16 and 4 leaf samples, respectively. RT-PCR test results revealed that PPV was found in none of the sweet cherry leaf samples. As a result of this study were obtained the infection rate % 10.13 of PPV, while PNRSV had an infection rate as % of 2.53. The total infection rate in the Tekirdag province for both viruses were found % 12.66.
