Özet
Bu çalışmada, Trakya bölgesinde yetişen Solanum dulcamara (Solanaceae) bitkisinin ham ekstrelerinde antihiperglisemik (antidiabetik) aktivite tayini ile fitokimyasal açıdan incelenmesi, sekonder metabolitlerinin izolasyonu, moleküler yapılarının açıklanması amaçlanmıştır. Çalışma kapsamında çalışılan Solanum dulcamara bitkisi, Trakya bölgesinde Temmuz-Ağustos 2012'de, Edirne (Söğütlük) civarından toplanmıştır. S. dulcamara bitkisinin meyve ve yaprakları, n-hekzan, diklorometan, etil asetat ve metanol ile polarite artış sırasına göre oda sıcaklığında maserasyon yöntemine göre ekstrakte edilmiştir. S. dulcamara'nın yaprak ve meyvelerinden elde edilen metanol ekstreleri, diabeti olan sıçanlar üzerine uygulanarak, ekstrelerin antihiperglisemik etkisi araştırılmıştır. Çalışma sonuçlarına göre, meyve ve yaprak ekstrelerinin hiperglisemi üzerine olumlu etkilerinin olmadığı tespit edilmiştir. Bu çalışmada hem meyve hem de yaprak ekstreleri, silikajel dolgu maddeleri içeren kolonlarda, ilk ayırıma tabi tutulmuştur. Buradan elde edilen fraksiyonlara ince tabaka kromatografisi (TLC) uygulanarak, benzer fraksiyonlar birleştirilmiştir. TLC sonucunda, belirlenen alt fraksiyonlar, ters faz kolonlu preperatif HPLC'de saflaştırılmıştır. İzole edilen saf maddelerin yapıları, 1D, 2D NMR (1H-1H COSY, DEPT, HMBC, HSQC) teknikleri, IR ve ESI-MS kullanılarak kimyasal yapıları açıklanmıştır. Bu çalışmanın sonucunda, S. dulcamara'nın yaprak ve meyve eksteresinden, bir biflavonoid (6,2',3'',5'',4'''-pentahidroksi-3,7''-biflavon), üç withanolide, bir steroid bileşiği olmak üzere toplam 5 bileşik izole edilmiştir. Bunlardan, biflavon bileşiği ise, doğal ürünlerden ilk kez izole edilen, yeni bir biflavonoid bileşiği olarak literatüre geçecektir.
The aim of this work was the isolation and structure elucidation of flavonoids, and other secondary metabolites from Solanum dulcamara (Solonaceae) and investigation of its antioxidant activities. Plants were collected in their flowering time (July and August 2012) in Edirne (Söğütlük) later it were separated, fruit and leaves. The air dried plant parts (fruit and leaves) were macerated with n-hekzane in room temperature. The solvent will be evaporated under vacuum. The resudie were be extracted by diethylether, ethyl acetate and metanol, respectively. The methanol extracts of leaves and fruits of S. dulcamara were investigated for the antihyperglycemic effect on the diabetic rats. As a result of this study, fruit and leaf extracts have been found not to have beneficial effects on hyperglycemia. In this work, the methanol extracts of leaves and fruits of S. dulcamara were subjected on silicagel/polyamid column and polar subfractions will be applied to the preperative RP-HPLC for the purifications of the polar compounds. For less polar compounds small sized column choromatography were used. Structure elucidation of the purified compounds were carried out by means of spectroscopic techniques such as 1D and 2D NMR (1H-1H COSY, DEPT, HMBC, HSQC) techniques, IR and ESIMS. As the result of this study, it was isolated a biflavonid (6,2',3'',5'',4'''pentahydroxy-3,7''-biflavon), three withanolides and a streoid compound from the fruit and leave extract of S. dulcamara. The biflavone compound was isolated from natural products for the first time in this study.
