Özet
Propolis bal arılarının bitkilerden topladıkları çeşitli ürünleri kullanarak ürettikleri reçinemsi doğal bir maddedir. Propolis eski çağlardan beri başta sağlık olmak üzere diğer alanlarda da yaygın bir şekilde kullanılmaktadır. Bilinen biyolojik aktivitelerinden bazıları; antibakteriyel, antiseptik, antitümoral, antiinflamatuar, antioksidan ve anestezik özellik göstermesidir. Hücre döngüsü, hücre büyümesi ve bölünmesinin kontrol edildiği evreleri içeren bir döngüdür. Hücrelerin döngüsünün kontrolü doku homeostazı ve kanser açısından önemlidir. Bu çalışmada; Türk propolisinin HEK293T ve HeLa hücrelerinde hücre döngüsüne etkisinin araştırılması amaçlanmaktadır. Çalışmamızda HEK293T ve HeLa hücreleri Türkiye'de üretilen ticari formdaki iki farklı propolisin LD50 konsantrasyonları ile in vitro koşullarda 24 saat süreyle muamele edilerek hücre döngüsü analizleri gerçekleştirilmiştir. Akım sitometri ile yapılan hücre döngüsü analizleri sonucunda; Propolis 1'in G1 fazındaki hücre sayısını kontrole kıyasla, HeLa hücrelerinde %24 oranında (p=0,004), HEK293T hücrelerinde %27,4 oranında (p=0,024) istatistiksel olarak anlamlı düzeyde düşürdüğü saptanmıştır. Propolis-2'nin ise kontrole kıyasla HEK293T ve HeLa hücrelerinde hücre döngüsünün herhangi bir evresindeki hücre yüzdesini istatistiksel olarak anlamlı bir düzeyde etkilemediği saptanmıştır. Çalışmamızdan elde edilen deneysel veriler Propolis-1'in HEK293T ve HeLa hücrelerinin hücre döngüsünün S-G2/M evrelerinde birikmesine sebep olduğunu ve hücre döngüsünü durdurduğu göstermektedir.
Propolis is a resinous substance produced by honey bees using the clothes they collect from their plants. Since ancient times, propolis has been used as a supplement in the treatment of many diseases and other areas. Antibacterial, antiseptic, antitumoral, anti-inflammatory, antioxidant and anesthetic effects are some of its biological activities that are known and researched. The cell cycle is a cycle that includes phases in which cell growth and division are controlled. Control of the cell cycle is important for tissue homeostasis and cancer. In this study, it was aimed to investigate the relationship between propolis activity and cell cycle in HeLa and HEK293T cells. In our study, cell cycle analyzes were performed by 24 hours in vitro treating HEK293T and HeLa cells with LD50 concentrations of two different commercial forms of propolis produced in Turkey. As a result of cell cycle analysis with flow cytometry; It was determined that propolis 1 statistically significantly reduced the number of cells in the G1 phase by 24% (p=0.004) in HeLa cells and 27.4% in HEK293T cells (p=0.024) compared to the control. It was determined that propolis-2 did not affect the percentage of cells at any stage of the cell cycle in a statistically significant level in HEK293T and HeLa cells compared to the control. Experimental data obtained from our study show that Propolis-1 causes HEK293T and HeLa cells to accumulate in the S-G2/M phases of the cell cycle and arrets the cell cycle.
