Domates bakteriyel hastalık etmelerine karşı yeni tip kimyasal formülasyonların antibakteriyel etkinliklerinin belirlenmesi

Özet
Domates yetiştiriciliğinde önemli bakteriyel hastalık etmenlerinden olan Pseudomonas syringae pv. tomato (Pst), Xanthomonas euvesicatoria (Xeu) izolatları bitkide bakteriyel benek ve bakteriyel leke oluşturarak ürün kalitesinde bozulmaya ve veriminde ciddi kayıplara neden olurlar. Domates bitkisinde bu hastalık etmenleri ile mücadelede kullanılabilecek nitelikte etkili ruhsatlı anti bakteriyel ürünler bulunmamaktadır. Bu çalışmada; Domates (Solanum lycopersicum L.) tarım alanlarında önemli hastalık etmenlerinden olan Pst ve Xeu izolatları üzerinde antimikrobiyal etki sağlayabilecek yeni tip formüllerin oluşturulması ve bu formüllerin hedef mikroorganizmalar üzerindeki antimikrobiyal etkinlikleri araştırılmıştır. Bu amaçla seçilmiş olan aktiflerin etkili oldukları dozlar in vitro koşullarda MIC metodu ile belirlenmiştir. Hedef mikroorganizmalar MALDI-TOF MS cihazı ile tanımlanmıştır. Etkinlikleri MIC metodu ile belirlenen aktifler ile in vitro koşullarda formülasyonlar oluşturulmuştur. Formüllerin etkinlikleri DDM metodu ile Pst ve Xeu izolatları üzerinde in vitro koşullarda her formül üç tekrarlı ve pozitif kontrol olarak kullanılan antibiyotik disk ile kıyaslanarak test edilmiştir. Formülasyonların etkinlikleri petrilerde oluşturdukları temiz alanlar ölçülerek hesaplanmıştır. Bu çalışma neticesinde oluşturulan 10 farklı formülasyonun tamamının Pst ve Xeu izolatları üzerinde (8-30 mm) arasında etkinlik gösterdiği saptanmıştır. Elde edilen test sonuçları göz önünde bulundurulduğunda bu formüllerin hedef mikroorganizmalara karşı kimyasal mücadelede kullanılabileceğini belirlenmiştir. Bu formülasyonların tarımsal alanlarda kullanılabilmesi için in vivo denemeler yapılmalı, toprak ve bitki bünyesindeki bozunumları incelenmelidir.
Pseudomonas syringae pv. tomato (Pst), Xanthomonas euvesicatoria (Xeu) isolates, which are important bacterial disease factors in tomato cultivation, cause bacterial blemish and bacterial spots on the plant, causing product quality deterioration and serious losses in yield.There are no effective, licensed antibacterial products that can be used to combat these disease agents in tomato plants.In this study; The creation of new types of formulas which can provide antimicrobial effect on Pst and Xeu isolates, which are important disease agents in Tomato (Solanum lycopersicum L.) agricultural areas, and the antimicrobial activities of these formulas on target microorganisms were investigated. The effective doses of the selected actives for this purpose were determined by the Minimum Inhibition Concentration Method (MIC) under in vitro conditions. Target microorganisms were identified by MALDI-TOF MS. Formulations were formed with the actives whose activities were determined by the (MIC) Method under in vitro conditions. The efficacy of the formulations was tested by Disk Diffusion Method (DDM) on Pst and Xeu isolates under in vitro conditions. In triplicate each formulation was compared with the antibiotic disk used as a positive control. The efficacy of the formulations was calculated by measuring the clean areas they created in petri dishes. As a result of this study, it was determined that all 10 different formulations were effective on, Pst and Xeu isolates (8-30 mm). Considering the test results obtained, it was determined that these formulas could be used in chemical control against target microorganisms. In order for these formulations to be used in agricultural areas, in vivo trials should be conducted and their degradation in soil and plant tissue should be examined.
