Öğe Characterization and Lidocaine Release Behavior of Chitosan/ Sodium Alginate/ Clinoptiolite Nanocomposite Hydrogel(Turkish Chemical Society, 2024) Sözkes, Sarkis; Taşdelen, Betül; Erdoğan, Sevil; Gülenç, Nadide Gülşah; Koruyucu, AslıhanIn this study, the novel nanocomposites were prepared from the natural biopolymers, chitosan (CS), sodium alginate (SA) and clinoptiolite (CL) particles, and also having glutaraldehyde as a crosslinker by cryogelation technique. CS biopolymer was produced from crayfish Astacus leptodactylus Eschscholtz, 1823. Characterization of the prepared CS, CS-co-SA (CS/SA) and drug loaded CS/SA/ CL nanocomposite were performed by Fourier Transform Infrared Spectroscopy (FTIR) and Scanning Electron Microscopy (SEM) analyses. The anesthetic drug release behavior of the prepared nanocomposite was investigated for the model drug lidocaine (LD) using UV-Vis spectrophotometry and High Performance Liquid Chromatography (HPLC) techniques. The effect of different LD and CL content on the drug release behavior of the prepared nanocomposite were studied. LD release data was fitted to various kinetic models to study the drug release behavior. The LD release from all the prepared nanocomposite hydrogels fitted zero-order, first-order, Higuchi, and Korsmeyer-Peppas models. The swelling and drug release properties of the new CS-based nanocomposite hydrogels were improved with the inclusion of SA and CL in the gel structure. © 2024, Turkish Chemical Society. All rights reserved.Öğe Conducting hydrogels based on semi-interpenetrating networks of polyaniline in poly(acrylamide-co-itaconic acid) matrix: synthesis and characterization(Wiley, 2017) Taşdelen, BetülIn this work, acrylamide/itaconic acid copolymeric hydrogels are prepared by free radical polymerization initiated by redox initiators of potassium persulfate and N,N,N,N-tetramethyl ethylene diamine; N,Nmethylene bisacrylamide was employed as a crosslinking agent. Aniline monomer was absorbed in the network of poly(acrylamide-co-itaconic acid) P(AAm-co-IA) hydrogel and followed by gamma radiation induced polymerization at room temperature. The novel semi-interpenetrating network was comprised of linear polyaniline immersed in P(AAm-co-IA) matrix. Electrical conductivity of the hydrogels was measured using four-probe technique. The conductivities for the prepared hydrogels are found to increase from 5.5x10(-7)Scm(-1) for P(AAm-co-IA) alone to 4.4x10(-3)Scm(-1) for semi-interpenetrating polymer network P(AAm-co-IA)/polyaniline. Thus, a new composite hydrogel with good conductive properties also displaying enhanced mechanical strength and pH sensitivity was prepared. Sentezlenen poliakrilamid ile graftlanmış jelatin (PA-g-GA), CL içerikli PA-g-GA ve ilaç yüklü PA-g-GA/CL kompozit hidrojellerinin karakterizasyonu FTIR ve SEM kullanılarak yapıldı. Sentezlenen yenkompozit hidrojelin ilaç salım davranışı, UV-Vis spektrofotometri ile incelendi. Farklı CL miktarı ve farklı ilaç konsantrasyon içeriklerinde sentezlenen yeni tip nanokompozit hidrojellerin, şişme ve ilaç salım davranışları incelendi. FTIR ve SEM analizleri, PA-g-GA/CL kompozit hidrojelinin başarıyla sentezlendiğini ve LD ilacıyla yüklendiğini ortaya koydu. Kompozitteki ilaç yükleme ve CL miktarı arttıkça LD salınımı (%) azalmıştır. İlaç salınım davranışını incelemek için LD salınım verilerine çeşitli kinetik modellere uyarlandı. Hazırlanan tüm hidrojellerin ilaç salımı Korsmeyer-Peppas modeline uymaktadır. Yeni nanokompozitlerin şişme ve ilaç salım özellikleri, jel yapısına klinoptiolitin eklenmesiyle geliştirildi.Öğe Preparation and characterization of chitosan/AMPS/kaolinite composite hydrogels for adsorption of methylene blue(Springer Science and Business Media Deutschland GmbH, 2021) Taşdelen, Betül; Çifçi, Deniz İzlen; Meriç, SüreyyaThis study aims to evaluate a new synthesized composite hydrogel prepared by gamma radiation polymerization of chitosan (CS) and 2-acrylamido-2-methylpropane sulfonic acid (AMPS) with kaolinite. The enhancement of kaolinite on the swelling, diffusion, and adsorption properties of CS/AMPS/Kaolinite hydrogels was studied using methylene blue (MB) as model compound. Effect of chloride ion on swelling and MB adsorption was also studied. A non Fickian behavior was observed for diffusion of water and MB into hydrogels. Adsorption capacities of the hydrogels significantly improved to be 52.9 mg/g, 39.7 mg/g, 30.7 mg/g, and 21.7 mg/g dry gel for CS/AMPS/Kaolinite-2, CS/AMPS/Kaolinite-1, CS/AMPS, and CS hydrogels, respectively by the addition of kaolinite. The equilibrium data for dye adsorption showed that Langmuir isotherm better fitted the MB adsorption with a high correlation (R2 > 0.99) than Freundlich, Temkin, and Elovich isotherms for all types of synthesized hydrogels. The chloride content affected the adsorption of MB in the order of AlCl3 > CaCl2 > NaCl. CS/AMPS/Kaolinite hydrogels can be used for adsorption of cationic dyes such as MB as an effective and promising adsorbent. It is to be emphasized that the addition of AMPS and kaolinite in the gel structure improved swelling and adsorption capability of the new composite hydrogels. © 2021, The Author(s), under exclusive licence to Springer-Verlag GmbH Germany, part of Springer Nature.Öğe Preparation and characterization of chitosan/hyaluronic acid/itaconic acid hydrogel composite to remove manganese in aqueous solution(Desalination Publications, 2021) Taşdelen, Betül; Çifçi, Deniz İzlen; Meriç, SüreyyaIn this work, new hydrogel composites composed of chitosan (CS)/hyaluronic acid (HA)/itaconic acid (IA) were synthesized by gamma radiation. Triple combination was prepared using gamma rays (25 kGy) at ambient temperature through simultaneous free radical polymerization and crosslinking. The swelling and diffusion properties of the hydrogels were investigated before submission to the adsorption experiment for removal of manganese (Mn) in aqueous solutions. The equilibrium mass percentage swelling of CS/HA hydrogels increased from 2,400 to 7,786 as the addition of IA in the CS/HA formulation. Fourier transform infrared and scanning electron microscopy were used for the surface characterization and chemical structure of the hydrogels. Mn uptake and release properties of the newly prepared composites improved with the including IA in the gel formulation of the CS/HA hydrogels. Adsorption capacity of Mn increased from 15.46 to 18.23 mg/g with the inclusion of IA in the formulation of CS/HA. It was found that adsorption capacities of Mn by both gels were competitive with some of the CS based composites used in Mn adsorption studies. © 2021 Desalination Publications. Aniline monomer was absorbed in the network of PAAm/Kaolin composite hydrogel and followed using free radical solution polymerization at room temperature. The novel semi-interpenetrating network was comprised of linear polyaniline immersed in PAAm/Kaolin composite. Electrical conductivity of the hydrogels was measured using four-probe technique. The conductivity for the prepared new semi-interpenetrating polymer network (s-IPN) is found to be 1.1x10(-4) S.cm(-1). The s-IPN hydrogel systems were employed for swelling experiments in water. The drug uptake and release properties of the prepared hydrogels were also studied. Methylene Blue (MB) was selected as a model drug. The incorporation of Kaolin and PAN lead to an increase in electrostatic interaction between charge sites on carboxylate ions and cationic MB molecules. It is to be emphasized that the addition of Kaolin and PAN in the gel structure improved mechanical and electrical properties and the adsorption capability of the new composite hydrogels. The stimuli responsiveness, swelling properties and mechanical properties of NIPAAm/IA//Pumice composite hydrogels were investigated. The parameters of swelling and diffusion of water in dye solutions were evaluated for the composite hydrogels. Water and Methylene Blue (MB) dye diffusion into hydrogels was found to be non-Fickian in character. Highly swollen NIPAAm/IA/Pumice composite hydrogels were used for sorption of water soluble cationic Methylene blue (MB) dye. MB adsorption capacities of the hydrogels were found to be 8.95,17.67 and 22.62 mg MB/g dry gel in PNIPAAm, P(NIPAAm/IA) and P(NIPAAm/IA/Pumice) systems, respectively. It is to be emphasized that the addition of Pumice in the gel structure improved mechanical properties and the adsorption capability of the new composite hydrogels. (C) 2016 Elsevier B.V. The use of the hydrogel samples as a drug delivery system was investigated by an anticancer drug. 5-Fluorouracil (5-FU) was used as a model anticancer drug to investigate the drug uptake and release kinetics of hydrogels. The properties of the hydrogels were evaluated in terms of swelling, drug uptake and release behaviours. Chemotherapy is an indispensable treatment method in many cancer treatments. Hydrogels have the ability to absorb/desorb water thanks to their cross-links and 3-dimensional network structures. In order to eliminate the disadvantages of chemotherapy applications and to ensure that a sufficient dose of drug is given to the patient, targeted drug release technologies have been developed and hydrogels have come to the fore in this field. Natural polymeric hydrogels are widely used in the medical field. Chitosan(CS) natural biopolymer has been used in controlled drug release systems for many years due to its unique properties such as high biocompatibility, biodegradability and low toxicity. By using different natural biopolymers with CS, the swelling and drup uptake capacity properties of the hydrogels can be improved. In this article, the swelling and drug uptake capacity of CS, CS/HA and CS/HA/GEL hydrogels in the 5-Fluorouracil (5-FU) used as a model drug was evaluated and drug uptake capacities were determined by UV-Vis spectrophotometer. As a result, swelling and drug uptake capacities of CS, CS/HA and CS/HA/GEL hydrogels in anticancer drug solution were investigated. While the 5-FU retention capacity (1.2 mg 5-FU/g dry gel) increased with the addition of HA to CS, a decrease in 5-FU retention capacity (0.4 mg 5-FU/g dry gel) was observed with the addition of GEL to CS. All the prepared hydrogels are promising in future for controlled drug delivery sytems in biomedical applications.Öğe Synthesis and Characterization of Natural Polymer Based Hydrogels(Tekirdağ Namık Kemal Üniversitesi, 2023) Güder, Ömer Faruk; Taşdelen, Betül; Akyol, UğurSoft tissue expanders, which have many uses in biomedical applications, are also encountered in surgical applications. Soft tissue expanders play an important role in ensuring osseointegration of dental implants. In this study, natural polymers chitosan (CS), hyaluronic acid (HA) and gelatin (GEL) were used. Synthesis of CS, CS-HA, CS-GEL, GEL-HA and CS-HA-GEL hydrogels has been performed. Gamma radiation was used in hydrogel synthesis with HAP. They were characterized by Fourier Transform Infrared Spectroscopy (FTIR) and Scanning Electron Microscopy (SEM). Dynamic swelling tests were performed in distilled water. Swelling behavior and swelling kinetics of hydrogels were determined. The effect of cross-linker concentration and different cross-linkers (glutaraldehyde (GA) and hydroxyapatite (HAP)) on swelling behavior was evaluated. Fickian mechanism has been observed in the diffusion of hydrogels. The results show that the hydrophilic CS-HA-1 hydrogel with the highest mass swelling and the most suitable swelling profile can be used as a soft tissue expander in biomedical applications.Öğe Tıbbi Cihazların Sterilizasyonunun Proses Validasyonu için Hazırlanan Boya-Akrilamid/İtakonik Asit Jel Dozimetresi(2021) Taşdelen, Betül; Sözkes, SarkisAmaç: Bu çalışmada tıbbi cihazların sterilizasyonunun proses validasyonu için kullanılacak yeni bir polimer jel dozimetre (PGD), gama ışınları kullanılarak hazırlanmıştır. İtakonik asit, akrilamid, jelatin, farklı boyalar [metilen mavisi, metil turuncusu ve kristal mor (KM)] ve oksijen tutucu olarak N, N’-metilenbisakrilamid ve askorbik asitten oluşan yeni polimerik sistemler üzerinde çalışılmıştır.Gereç ve Yöntem: Hazırlanan yeni PGD dozimetre formülasyonlarındaki farklı boyaların doz hız sabitleri ve radyasyon kimyasal verimleri üzerindeki etkisi, birçok tıbbi cihaz, farmasötik ürün ve biyolojik dokunun ultraviyole kullanılarak sterilizasyonu için tipik olan 7-28 kGy yüksek doz aralığında spektrofotometre yöntemi ile araştırılmıştır.Bulgular: Hazırlanan PGD dozimetrelerinin spektrumu, numunelerin gama ışıması üzerine bir değişikliğe uğraması ve mavi jel renginin ağartılması,radyasyon dozu ile artar; tıbbi cihazın sterilizasyon dozunda (25 kGy) bile, ışınlanmamış KM-PGD’nin koyu mavi rengi neredeyse renksiz olana dönmüştür. Işınlanmamış ve ışınlanmış PGD dozimetrelerinin tepkisi, 60 günlük saklama süresi boyunca oldukça kararlıdır.Sonuç: Hazırlanan boya-PGD dozimetrelerinin yüksek doz gerektiren radyasyon proses uygulamalarında, örneğin; tıbbi cihazlar ve sağlık bakım ürünlerinin sterilizasyonunun proses validasyonunda faydalı olacağı kanaatindeyiz.
