Kıvırcık koyunlarında beta laktoglobulin polimorfizminin PCR-RFLP yöntemiyle belirlenmesi

Abstract

Bu tezin amacı yerli koyun ırklarımızdan biri olan saf Kıvırcık koyununda ?- laktoglobulin polimorfizminin belirlenmesidir. Bu amaçla yerli koyun ırklarımızdan ve saf olduğu bilinen 48 baş Kıvırcık koyun kullanılmıştır. Çalışma PCR-RFLP tekniği kullanılarak gerçekleştirilmiştir. Ayrıca araştırmada bu ırk için ?-laktoglobulinin iki farklı alleline ait ( A ve B) ile üç farklı genotip (AA, AB ve BB) belirlenmiştir. Bu gen lokusuna ilişkin allel gen frekansları ise ?-Lg A ve B için 0.468 ve 0.532 olarak hesaplanmıştır. Sürünün, HardyWeinberg yasasına göre genetik dengede olduğu bulunmuştur. Bu sayede Kıvırcık koyun ırkının bu özellik için genotipik yapısı belirlenmiştir.

The aim of this study is to determine the genetic polymorphism of the ?-laktoglobulin gene in Kıvırcık pure breed. 48 pure breed that isone of native sheep breeds of Turkey was used for this study . The study was carried out by means of PCR-RFLP technics. Also, two genetic variants (A and B) and three genotypes (AA, AB and BB) were determined for this breed. The gene frequencies of ?-Lg A and B were calculated as 0.468 and 0.532 in Kıvırcık. The populations of this breed was in equilibrium according to Hardy-Weinberg rule. The genetic structure of this breed was exemined in the present study revealed polymorphism at ?-laktoglobulin gene locus.
