Sıvı azotta depolanmış Ephestia kuehniella Zeller yumurtalarını kullanarak Trichogramma pintoi Voegele’nin kitle üretimi

Abstract

Trichogramma’ların kitle üretiminde konukçu yumurtasının sıvı azot içinde depolanması, alternatif bir yöntem olabilir. Bu çalışmada sıvı nitrojende 1, 2, 3, 4, 5 ve 6 hafta boyunca depolanan Ephestia kuehniella Zeller yumurtalarından elde edilen Trichogramma pintoi Voegele’nin parazitleme performansı ve dişi ömür uzunluğu incelenmiştir. Depolama sonrasında yumurtaların çözünmesi için iki farklı yöntem uygulanmıştır. İlk olarak yumurtalar tanktan çıkarıldıktan sonra 24 saat buzdolabında bekletilmiş ve sonrasında 50 °C su banyosunda 30 dakika tutulmuştur. İkincisinde ise yumurtalar tanktan alındıktan sonra 24 saat boyunca sadece buzdolabında bekletilmiştir. Her iki yöntemin uygulanması ile sonuçlar arasında önemli bir farklılık olmadığı tespit edilmiştir. En yüksek parazitlenme bir ve iki hafta depolanan yumurtalardan sağlanmıştır.

The storage of host eggs in liquid nitrogen may represent an alternative method in the mass production of Trichogramma.
