Trakya Bölgesi doğal florasında bulunan bazı yabani fiğ (Vicia sp.) türlerinin kromozom sayıları ve karyotip karakterizasyonunun belirlenmesi

Abstract

Bu çalışma, Trakya Bölgesi'nde doğal olarak yetişen Vicia cinsine ait V. pannonica subsp. pannonica, V. sativa subsp. nigra, V. villosa subsp. villosa, V. lutea subsp. hirta, V. hybrida L. türleri olmak üzere 7 farklı genotipin kromozom sayıları ve karyolojik özelliklerinin belirlenmesi amacıyla yürütülmüştür. Sitolojik karakterizasyon çalışması Feulgen boyası kullanılarak gerçekleştirilmiştir. Çalışmada türlere ait karyotip analizi yapılarak idiogramları çizilmiştir. Çalışma sonucunda V. sativa subsp. nigra kromozom sayısı 2n=14, V. pannonica subsp. pannonica kromozom sayısı 2n=14, V. lutea subsp. hirta kromozom sayısı 2n=14, V. villosa subsp. villosa kromozom sayısı 2n=14, Vicia hybrida L. kromozom sayısı 2n=12 olarak tespit edilmiştir. Çalışmada Vicia cinsinde yer alan genotiplerin karyotip formülleri; Vicia sativa subsp. nigra türü 21-2 nolu genotipte 4Sm + 3M olarak, Vicia pannonica subsp. pannonica türü 15-14 nolu genotipte 4St + 2Sm, Vicia lutea subsp. hirta türü 7-6 nolu genotipte 7Sm, 15-I-24 nolu genotipte 7St olarak, Vicia villosa subsp. villosa türü 15-I-43 nolu genotipte 6Sm + 1M, 120-3 nolu genotipte 7Sm, Vicia hybrida L. türü 22-1 nolu genotipte 5St + 1Sm olarak belirlenmiştir. Çalışılan genotipler arasında en küçük kromozom boyuna 2.391 µm ile Vicia sativa subsp. nigra türü 21-2 nolu genotip sahipken, en büyük kromozom boyuna ise 8.459 µm ile Vicia lutea subsp. hirta türü 15-I-24 nolu genotip sahiptir. Kol oranlarında ise en küçük kol oranı 1.500 µm ile Vicia villosa subsp. villosa türü 15-I-43 nolu genotip, en büyük kol oranına ise 4.754 µm ile Vicia hybrida L. türü 22-1 nolu genotipte rastlanmıştır.

This study was conducted to determine the chromosome numbers and karyological characteristics of 7 different genotypes, V. pannonica subsp. pannonica, V. sativa subsp. nigra, V. villosa subsp. villosa, V. lutea subsp. hirta, V. hybrida L. species, which were naturally grown in Thrace Region of Turkey. The cytological characterization study was performed using Feulgen stain. In the study, idiograms of the species were drawn by karyotype analysis. As a result of the study, chromosome numbers of the species were determined as V. sativa subsp. nigra 2n=14, V. pannonica subsp. pannonica 2n=12, V. lutea subsp. hirta 2n=14, V. villosa subsp. villosa 2n=14, Vicia hybrida L. 2n=12, respectively. In the study, the karyotype formulas of the genotypes in the genus Vicia were determined; Vicia sativa subsp. nigra species as 4Sm + 3M in genotype 21-2, Vicia pannonica subsp. pannonica species as 4St + 2Sm in genotype 15-14, Vicia lutea subsp. hirta species as 7Sm in genotype 7-6, 7St in genotype 15-I-24, Vicia villosa subsp. villosa species as 6Sm + 1M in genotype 15-I-43, 7Sm in genotype 120-3, Vicia hybrida L. species 5St in genotype 22-1 as +1Sm. Among the genotypes studied, the shortest chromosome length was determined at Vicia sativa subsp. nigra genotype 21-2 with 2.391 µm, the longest chromosome was determined at Vicia lutea subsp. hirta genotype 15-I-24 with 8.459 µm. The lowest arm ratio has been encountered at Vicia villosa subsp. villosa genotype 15-I-43 with 1.500 µm and the highest at Vicia hybrida L. genotype 22-1 with 4.754 µm.
